   Tesofensine is a serotonin-norepinephrine-dopamine reuptake inhibitor originally developed for the treatment of Parkinson's disease and Alzheimer's disease. According to a report by researchers at the 16th European Congress on Obesity, tesofensine helps obese patients lose weight and reduces weight in the appropriate parts of their bodies and the appropriate components (fat).
   Danish researchers studied the effects of tesofensine treatment on body composition, including body weight, fat mass, waist circumference, and sagittal diameter (upper abdominal size). The study was part of a randomized, double-blind, placebo-controlled Phase IIb clinical trial involving 203 obese men and women with a body mass index between 30 and 40 (with no other diseases). After a 2-week run-in period of dietary control and physical exercise, they were divided into groups for 4 weeks of drug treatment, receiving daily doses of 0.25mg, 0.5mg, or 1.0mg of tesofensine or a corresponding placebo. The results showed that tesofensine is highly effective against central obesity.
   Another study indicated that low-dose tesofensine is safe and effective for treating obesity. A dose of 0.5mg of tesofensine produced significant weight loss effects with only mild and transient adverse reactions. These conclusions were drawn from a research report presented at the annual conference. The lead author, Dr. Arne V. Astrup, Head of the Department of Nutrition at the Faculty of Life Sciences, University of Copenhagen, reported that patients receiving 0.25mg of tesofensine experienced an average weight loss of 4.54%, the 0.50mg group lost 9.18%, and the 1.0mg group lost 10.6%. Tesofensine caused an increase in blood pressure and heart rate, proportional to the dose. Adverse reactions were generally mild and temporary. Dr. Astrup suggested that 0.5mg should be the optimal dose for tesofensine. He saw no reason to use the 1.0mg dose, as it did not result in greater weight loss but increased adverse reactions.
