The XenoWorks? Digital Microinjector is primarily used for transgenic mammalian workstations and injections of adherent cells, C. elegans, Drosophila, zebrafish, Xenopus, and sea urchins. It can accommodate oocytes and embryos while providing separate pressure channels for pronuclear microinjection of DNA or gentle control of embryonic stem cell transfer into blastocysts. The microinjector employs two independent pressure channels: a holding channel and an injection/transfer channel, controlled via a remote user interface.

The holding port is a separate vacuum channel designed for gently suspending cells and can be toggled on or off as needed. A responsive rotary controller adjusts the pressure based on the desired level of retention.

The injector/transfer port is the second channel, capable of delivering high pressure for injections ranging from femtoliter to microliter volumes, as well as gentle positive/negative pressure for embryonic stem (ES) cell transfer. The transfer channel features five range settings to optimize the system for each application and user preference. It also includes a clearing function to address pipette clogging. This channel offers two injection modes: continuous or pulsed, combined with balanced pressure. In continuous mode, injection pressure is applied to the micropipette tip as long as the manual or foot switch is held. In pulsed mode, injection pressure is applied for precisely timed durations, adjustable in 0.01-second increments. Injection duration and pressure can be easily selected using the rotary control on the remote interface. Injections are executed using the [Inject] key on the remote interface or foot switch.

Ergonomics remain a top priority. The remote interface unit provides access to all necessary keys and rotary controls for manipulating parameters across both pressure channels, while the separate compressor controller can be positioned away from the microscope to eliminate vibration.

Beyond pronuclear injection, the XenoWorks Digital Microinjector is suitable for various intracellular and extracellular applications, including microinjection of adherent and suspended cells.
